After graduating from Barker College, Johnstone-Burt auditioned for the National Institute of Dramatic Art (NIDA) at 18. He was informed that he was too young to attend, so he traveled instead. When Johnstone-Burt returned to Australia, he auditioned for NIDA again and was accepted. In 2009, he played Jack in The Importance of Being Earnest in his graduation play.

In the musical drama film Goddess, he co-starred with Magda Szubanski.

In the third season of Miss Fisher’s Murder Mysteries, Johnstone-Burt reprised his role as Hugh. He also co-starred with Dwayne Johnson and Kylie Minogue in the feature film San Andreas. In the Network Ten drama The Wrong Girl, Johnstone-Burt played Vincent, the brother of the lead character Lily, played by Jessica Marais.
